Kyohei Hamura was born in Tokyo, Japan in 1966, and he became a member of the Tojo-kai yakuza clan at a young age. Rising through the ranks of the Matsugane-gumi, the violent and egotistical Hamura allied with the bent cop Mitsuru Kuroiwa in 1998, utilizing Kuroiwa's skills to dispose of dead bodies and later to carry out hits. Hamura later became captain of the family, becoming increasingly powerful as the rank-and-file of the family came to be loyal to Hamura over Patriarch Mitsugu Matsugane. In 2017, Hamura hired the homeless man "Red Nose" to stage a robbery of the Matsugane HQ and catch the lone guard Masaharu Kaito unprepared. After Kaito agreed to let the robber make off with the money rather than resist and potentially endanger Toru Higashi (who was, unbeknownst to Red Nose and Hamura, in the office, shining Matsugane's shoes), Hamura demanded that Kaito be expelled from the family. Matsugane agreed, thus removing Matsugane's most powerful loyalist and allowing for Hamura to effectively take control of the clan with a reluctant Higashi at his side. Privately, Hamura deeply respected Matsugane, and he intended to make the family stable through using brute force to make money.

At the same time, Ministry of Health official Mitsuru Ishimatsu contacted Hamura and Kuroiwa for help with protecting the AD-9 drug as it neared approval for human usage. As the drug had severe defects and ADDC vice-chair Toru Hashiki attempted to reveal its side effects (with the ultimate goal of helping the Kyorei-kai close down the ADDC and redevelop its land), Ishimatsu paid Hamura and Kuroiwa to get rid of Hashiki. Hamura reluctantly agreed, as he wanted to keep the Matsugane Family afloat with Ishimatsu's illicit money. Through Hamura, Kuroiwa was ordered by the ADDC to kill several men who posed a threat to both the drug and the institution, including three Kyorei-kai yakuza who had moved into Tokyo to establish a foothold for the Kyorei-kai's planned takeover.

Hamura was arrested for the murder of one of the Kyorei men, Toshiro Kume, following an altercation between himself, Kume, and Kume's boss Akira Murase at Club Amour in December 2018. Hamura's acquaintance Takayuki Yagami, a friend of Kaito's, was hired to help the lawyer Masamichi Shintani overturn the charges, and Yagami proved to the court that Hamura had been at a sauna at the time of Kume's murder. Following his acquittal, Hamura warned Yagami not to dig deeper into the case. When Yagami refused to drop the case, Hamura and his men resorted to beating down Yagami at Cafe Alps. After Yagami escaped with the help of the masked vigilante Fumiya Sugiura, Hamura ordered a manhunt for Yagami, sending Higashi to raid Yagami's office to intimidate him.

Yagami soon grew suspicious that Hamura was in league with the true murderer of the Kyorei Clan men, and his suspicions were confirmed when the Kyorei-kai shot a Matsugane-gumi member, Masakazu Kurimoto, who was mistaken for Hamura. Yagami eventually tracked Hamura down to a secret cho-han parlor in the Golden Gai neighborhood, but members of the Matsugane-gumi set the parlor on fire before rescuing their captain. Yagami lured Hamura out after visiting various Matsugane-gumi establishments to make his survival known. Hamura kidnapped Kaito as a result, but Yagami infiltrated Matsugane HQ to rescue him. Yagami proceeded to rescue Kaito, and Matsugane helped his two proteges interrogate Hamura at Honmaruen, where Hamura revealed that "the Mole" had been a policeman before becoming a hitman. Hamura also confirmed that all of the victims whose eyes were gouged out had been experimented on by Yoji Shono before dying.

Shortly after, the Matsugane-gumi raided Honmaruen to rescue Hamura, and Kuroiwa also arrived, attempting to kill Hamura to keep him quiet. However, Matsugane took a bullet to save Hamura, and he forgave Hamura's past misbehaviors before dying. Hamura, determined to avenge his boss and take revenge on the treacherous Kuroiwa, revealed to Yagami that "the Mole" was "Mitsuru Kuroiwa", and he then went into hiding before giving Yagami evidence that would incriminate Vice Minister of Health Kaoru Ichinose in the AD-9 conspiracy. He later testified at Kazuya Ayabe's trial, proving that Ayabe was not the murderer, but that Kuroiwa had carried out the AD-9-related homicides.
